VLC Twoflower x264 Profiles & Settings

Hello everyone,
I have been wandering around the forums and did some research on Google, but every information I have found is limited or too technical for me, so I will ask here and maybe someone can spare some time to answer:
1) The "x264 profile and level selection" is something that in fact affects all x264 videos played, is this statement correct?
2) Are the new profiles, for example the "high444 and high10" able to provide better output quality? If yes, which is better?
3) Is there a way to configure "x264 preset and tuning selection" and "Skip H.264 in-loop deblocking filter" so I can increase output quality? -> for some reason I have the feeling these can only decrease quality to improve performance.

And another thing I wanted to ask is; Twoflower does not save the option "Use system codecs if available, for WMV codecs". This is something it was properly saved in the previous versions, although if I remember correct it had a somewhat different name.
Is this a bug or just me?
